    Abstract:

    Inrecentdecades,thepopulationstructureofmanymarinefishhasdramaticallychangedundertheinfluenceofanthropogenicactivitiesandclimatechange.Tobetterunderstandthechangemechanism,DNAextractedfromformalin-fixedmarinefishspecimensisrequired.However,obtaininghighqualityDNAfromformalin-fixedspecimensisdifficult.Toobtainknowledgeforspecimensthatwerefixedin1962wereusedassamples,andanorthogonalexperimentwasdesignedfollowingtheL)table.TheDNAconcentrationwastakenastheevaluationindex.Thetissuetypes,anti-formaldehydetreatments,digestivetimesandextractionmethodsweretakenasselectionfactors.Thebestgroupwasdeterminedtobe℃anddryinginavacuumcentrifuge,3hofdigestionwithProteinaseKwithinawaterbathof55,andDNAextractionbykit.Anti-formaldehydetreatmentssignificantlyimprovedtheproductionofDNAandtheuseofvariousanti-formaldehydemethodsobtainedthebesteffect.Digestionaftera90DNAwasassessedbyPCRamplificationandsequencingthemethods
