    • Prokaryotic expression, antibody preparation, and identification of a cyclin B protein in the Chinese mitten crab Eriocheir sinensis and Malaysian giant prawn Macrobrachium rosenbergii

      Abstract:We amplified the open reading frames (ORFs) of the cyclin B gene from the ovarian cDNA of the Chinese mitten crab Macrobrachium rosenbergii. The ORFs from these two species were then ligated into the expression plasmids pGEX-2T and pET32a, respectively. Following transfer of the recombinant plasmids into a host bacteria, BL21 (DE3), the recombinant proteins were expressed as inclusion bodies by induction with IPTG. The optimum concentration of IPTG was 0.1 mmol/L for 4 h at 37C for GST-EsCB and 0.1 mmol/L PTG for 6 h at 30C for Trx-MrCB. The recombinant proteins were subsequently purified by affinity chromatography. The cyclin B antibody was further refined by immunizing New Zealand white rabbits with the purified protein. The highest resulting antiserum titer was 180 000 (ELISA). We verified the specificity of the antibodies against the recombinant protein and ovarian cyclin B protein by western blot. We isolated two isoforms of the cyclin B protein from the immature crab ovaries, but only one form from the prawn ovaries. Thus, our data suggest that the role of cyclin B in the regulation of oocyte maturation may differ between crabs and prawns. The cyclin B antibody will be useful for further study of the role of cyclin B on MPF activation and oocyte maturation.

    • Induction of CYP3A gene and enzyme activity expression in kidney cell of grass carp (Ctenopharyngodon idellus)

      Abstract:Cytochrome P450 3A (CYP3A) is the major cytochrome P450 responsible for the metabolism of endogenous and xenobiotic substrates.Unexpected drug–drug interactions in fish are generally associated with the induction of CYP3A activity, and can lead to the formation of drug residues, which threaten the safety of fishery products. We determined the ) on CYP3A mRNA expression and enzyme activity in a freshwater teleost, the grass carp (). Recently, a number of CYP3A genes have been isolated from several teleost species. Although CYP3A genes in fish contain multiple paralogs differing in gene expression pattern and tissue distribution, CYP3A enzymes exhibit similar catalytic properties because of their structural similarities. The CYP3A gene and its full-length cDNA of 1 849 bp were cloned from grass carp by RT-PCR. The CYP3A gene has an open reading frame (ORF) of 1 542 bp, a stop codon, a 3’-untranslated regions (3’ UTR), and a polyA signal is present in the 3’UTR region. The ORF encodes a 513 amino acid (aa) protein, which has a signal peptide (29aa), two transmembrane helixes (23aa and 23aa), a heme binding domain (HBD, 21aa), and six substrate recognition sites (SRS1-6). Multiple alignments showed that the CYP3A amino acid sequence from grass carp is 60% to 92% similar to other vertebrate CYP3As. CYP3A gene expression after induction by RIF in grass carp kidney cells (CIK) was assayed by quantitative real-time PCR. CYP3A-dependent erythromycin N-demethylase (ERND) activity was determined by spectrophotometry, and CYP3A activity was assessed by measuring the formation of formaldehyde. In the induced group, CYP3A mRNA expression reached a plateau at 8h, while the highest level of CYP3A activity occurred at 10h. The results indicated that CYP3A mRNA expression and enzyme activity were significantly higher than in the control group (0.05). The highest level of CYP3A mRNA expression appeared 2 hours before the maximum of enzyme activity. In addition, the transcription of level CYP3A was apparently higher than the enzyme activity, implying that induction by RIF affected enzyme activity by transcriptional regulation of the CYP3A gene.

Toxin-coregulated pilus (TCP) is a major virulence factor of As a major structural protein of TCP, toxin-coregulated pilin A (TcpA) is potential candidate for vaccine development. In this study, the diseased grass carp (gene fragment containing the open reading frame (ORF) was then subcloned into pGEX-4T-1 to construct expression plasmid pGEX-4T-1-tcpA. The recombinant fusion protein (rGST-TcpA) was expressed by IPTG induction incharacterization. Sequence analysis revealed that the ORF of thegene from the Y1 strain (GenBank accession no. EU649677) is 657 bp and encodes a protein of 224 amino acids. The gene is 99.6%–9% identical at the protein level to seven tcpA sequences in GenBank, which suggested that the TcpA protein is considerably conserved. SDS-PAGE analysis showed that the 47.0 kD rGST-TcpA fusion protein was mainly expressed in inclusion bodies. Western blotting demonstrated that rGST-TcpA could react specifically with mouse antisera raised against the pilus protein of strain Y1. The purified rGST-TcpA was used to immunize ). The titer of the antisera was 1:16, as determined by a double immunodiffusion test, and it could significantly inhibit adherence of Y1 strain to HEp-2 cells .       Abstract:relationship between transmission routes of AVNV and mesoplankton and microplankton in the ocean for culturing scallops. Plankton samples were collected in Liuqinghe Qingdao and Sanggou Bay, Rongcheng, from April to November 2009. In August, the AVNV copy number carried by plankton was identified by FQ-PCR. Results showed that plankton samples from both ocean areas carried the virus, and that the virus numbers differed with different particle sizes. Plankton samples with the same particle sizes at the Liuqinghe site carried more AVNV copy numbers than those at the Sanggou Bay site at a given time. Plankton particle sizes smaller than 200 mesh carried the greatest amount of viruses (viral DNA number reached 9.45×106 copy/mg DNA), followed by plankton particle sizes between 60−200 mesh size. Plankton particle sizes larger than 60 mesh carried the least amount viruses. Finally, results suggest that plankton can carry AVNV, and that they may be an important host of AVNV.

      Abstract:Tetra-primer amplification refractory mutation system-polymerase chain reaction (Tetra-primer ARMA-PCR) was introduced to investigate single nucleotide polymorphisms (SNPs) genotyping in Chinese shrimp () and 80 were studied. Twenty out of the 80 SNP tetra-primer ARMA-PCR primer sets were validated following touchdown profiles and the outer and the expected inner bands were amplified. Homozygous and heterozygous were detected by agarose gel and the genotypes were obtained. Polymorphism of these distinct loci was assessed using 30 individuals, and the results showed that the 20 loci were all polymorphic. The values of and PIC varied from 1.127 to 1.993, from 0.136 to 0.607, from 0.119 to 0.492 and from 0.145 to 0.3730, respectively. The results indicated that tetra-primer ARMA-PCR is a simple, rapid and efficient method for SNP genotyping which make it useful in a broad aspects of genetic and breeding studies.

      Abstract:We documented the penetration of sperm into the oocyte and the changes in nuclear behavior in using paraffin sections and fluorescent microscopy. The mature oocyte was egg-shaped, had an unbroken germinal vesicle, and remained in the prophase of the first maturation division. Sperm rapidly attached to the surface of the oocyte within 5 min of insemination, water temperature (21±1) °C. The sperm penetrated into the oocyte at a location close to vegetal pole triggering development of the fertilization membrane. The sperm nuclei then expanded and the fertilization membrane was completed within 25 min. The first and second maturation divisions were completed 10 and 50 min after insemination, and were accompanied by the exclusion of the first and the second polar bodies, respectively. The nuclei of the sperm and egg expanded 50–70 min later, respectively, and developed into the pronuclei. These then migrated to the center of the egg, forming an associated nucleus. The first cleavage was completed after 70–90 min, resulting in two daughter cells. The fertilization process lasted ~70 min, which is slightly longer than in most marine invertebrates. Our observations provide basic information for artificial breeding and the cultivation of new varieties of .

    • Estimation of parameters and breeding values for growth traits of largemouth bass

      Abstract:The genetic parameters and breeding values for growth traits (including body weight, body length, and body height) of the largemouth bass () were estimated using the best linear unbiased prediction (BLUP) method. 0.29±0.08 for body weight and 0.31±0.08 for body length; at six months of age, heritability was 0.28±0.10 for body weight, 0.26±0.10 for body length, and 0.29±0.10 for body height. These heritabilities indicated that there was a great potential for selective breeding in the growth traits of largemouth bass. At four and six months of age, the genetic correlations between body weight and body length were 0.750±0.082 and 0.790±0.094, respectively. At six months, the genetic correlation was 0.820±0.081 between body weight and body height and 0.990±0.001 between body length and body height. These genetic correlations indicated that selection for body weight would result in positive responses in other traits. orrelation analysis of different estimated breeding values (EBVs), at four and six months, gave correlation coefficients of 0.923 and 0.919 between comprehensive EBV and length EBV, and 0.914 and 0.890 between weight EBV and length EBV, respectively. These correlation coefficients showed that there was little difference in ranking for largemouth bass when using comprehensive EBV compared to one-trait EBV. The above parameters and breeding value results provide a theoretical basis and technical reference for the selective breeding for largemouth bass.

      Abstract:is an important component of the complement system, which interacts with collagen-like region of C1q and invokes many cell responses, involved in enhancing phagocytosis, promoting macrophage of microbial, inducing chemotaxis, and further enhancing the organism's immunity. In recent years, higher frequencies of disease epidemics have been reported. . Now we have successfully constructed cDNA library of ovarian tissue in , and identified PmC1qBP gene. To study the biological functions of PmC1qBP in , we compared the expression level of PmC1qBP in different tissues of female and male and development stages of ovary with semi-quantitative and real-time quantitative PCR. The results show that the expression level of PmC1qBP has significant differences between male and female in gonads, hemocytes, intestine and stomach. At the same time, the expression level of PmC1qBP is very low in nauplii and zoea larvae stage, which increases from mysis larvae stage and stabilizes during the maturation stages of ovary. The results reveal that PmC1qBP is an important functional gene during the ovary development of . The immune challenge experiment indicated that PmC1qBP is up-regulated in hepatopancreas after treated with LPS for 6 h. Penaeus monodon and might play an important function in immune regulation.
